La ciclina CNTD2: un nuevo inductor del desarrollo tumoral

Abstract
El cáncer colorrectal (CRC) es uno de los cánceres más comunes en todo el mundo, con un 8-10% de estos tumores que presentan una mutación BRAF (V600E). Las ciclinas son oncogenes comúnmente desregulados en muchos cánceres, pero el papel de la nueva subfamilia de ciclinas atípicas no ha sido caracterizada aún. En esta tesis se ha realizado un análisis sistemático de los niveles de expresión de proteínas de seis ciclinas atípicas en tumores de CRC humanos y varias líneas celulares, y encontramos que CNTD2 está significativamente sobreexpresado en los tejidos cancerígenos en comparación con el tejido sano adyacente. La sobreexpresión de CNTD2 en líneas celulares CRC aumenta su capacidad de proliferación y migración, así como varias propiedades mesenquimales, como la capacidad de formación de esferoides y el crecimiento independiente del anclaje. Curiosamente, CNTD2 aumenta el crecimiento tumoral in vivo en modelos xenógrafos de CRC con BRAF wt y disminuye el crecimiento tumoral en xenoinjertos con BRAF mutado, un hallazgo que se correlaciona con la importancia pronóstica de CNTD2 en pacientes con CRC. En consecuencia, el silenciamiento de CNTD2 disminuyó significativamente la proliferación de células BRAF CRC wt. Nuestros resultados sugieren que el resultado de la sobreexpresión de CNTD2 depende del contexto BRAF, y que CNTD2 puede representar un nuevo factor pronóstico y un objetivo de fármaco prometedor en el tratamiento de la CCR.
Además del CRC, el fenotipo agresivo promovido por CNTD2 se pone de manifiesto en su capacidad para inducir propiedades mesenquimales tan importantes para la producción de metástasis, también en otros cánceres como el de mama y pulmón. La sobreexpresión de CNTD2 aumentó la expresión proteica de varios marcadores de stemness y la capacidad de formación de oncosferas, que a su vez disminuyeron con el silenciamiento del mismo. Finalmente, el análisis del valor pronóstico de la expresión del gen CNTD2 en grandes conjuntos de pacientes con cáncer de mama, pulmón y CCR reveló una correlación significativa entre los altos niveles de expresión de CNTD2 y la reducción de la supervivencia global. En conclusión, nuestros resultados sugieren que CNTD2 es un nuevo inductor oncogénico, que representa un nuevo marcador pronóstico y diana terapéutica en cáncer.
