Abstract
Hoy en día la regeneración tisular mediante el uso de células madre mesenquimales (MSC) es un tema que está en estudio a nivel mundial. La cavidad bucal es un nicho interesante de obtención de mesenquimales. Las primeras en obtenerse fueron del tejido pulpar sano (DPSC). Sin embargo, hay poca investigación acerca de la capacidad inmunomodulatoria de las DPSC derivadas de pulpa sana, inflamada y de células pluripotentes (DPPSC). Frente al problema de investigación encontrado, surge la pregunta ¿cómo afecta la inflamación local la capacidad inmunomodulatoria de células madre mesenquimales obtenidas de pulpa dentaria humana? El objetivo general de esta tesis doctoral fue evaluar las características funcionales de las DPSC derivadas de pulpa sana e inflamada y determinar las propiedades inmunomoduladoras de las DPSC
En esta investigación se utilizaron células madre mesenquimales derivadas de pulpa sana y de pulpas con diganóstico de pulpitis irreversible; además utilizamos células derivadas de tejido gingival humano (GMSCs). Las DPSCs y las GMSCs fueron caracterizadas según los criterios establecidos por la “International Society for Cell Therapy” (ISCT). Para evaluar la capacidad inmunomoduladora, se midió el efecto de las MSC en la proliferación de los linfocitos (LT-CD3), con el fin de observar el efecto inmunoestimulador o inmunosupresor (aumento o disminución de la proliferación de linfocitos).
Los linfocitos fueron aislados de sangre periférica humana y activados con un mitógeno y marcados con un anticuerpo anti CD3. Los linfocitos fueron cocultivados con las MSC y se evaluó mediante citometría de flujo el porcentaje de proliferación de los LT-CD3 en las distintas condiciones experimentales ya sea en presencia y en ausencia de células.
