Rol de CPT1C en la supervivencia de las células madre mesenquimales

Abstract
La carnitina palmitoiltransferasa 1C (CPT1C) pertenece a la familia de enzimas CPT1, las cuales controlan la entrada de ácidos grasos al lumen de la mitocondria para su posterior oxidación. Sin embargo, la isoforma cerebral CPT1C se caracteriza, a diferencia de la A y la B, por tener una baja actividad catalítica, por disponer de 30 aminoácidos adicionales al final de la secuencia aminoacídica y por localizarse en el retículo endoplasmático. Recientemente, se ha demostrado que CPT1C también se expresa en células tumorales, confiriéndoles la capacidad de sobrevivir en situaciones de estrés metabólico, como son la ausencia de glucosa o la hipoxia.
Dada la alta similitud entre las células tumorales y las células madre, en este trabajo hemos querido estudiar si CPT1C se expresa en células madre adultas humanas y su hipotético rol protector en situación de estrés metabólico.
En primer lugar, demostramos que CPT1C se expresa en células madre mesenquimales (MSC) procedentes de la pulpa dental humana. En segundo lugar, aportamos que la sobre-expresión de CPT1C confiere supervivencia celular cuando disminuyen los niveles de glucosa o bien, cuando se inhibe la glucólisis con 2-desoxiglucosa y que el silenciamiento de CPT1C induce el efecto opuesto. En tercer lugar, demostramos que la sobre-expresión de CPT1C, en ausencia de glucosa, no altera la capacidad oxidativa de las mitocondrias pero sí aumenta los niveles de producción de ATP. Curiosamente, cuando las MSC no disponen de glucosa en el medio, aumentan los niveles de expresión de CPT1C endógena. Y para finalizar, este trabajo pone de manifiesto que dicha supervivencia celular se lleva a cabo a través de un aumento del ciclo autofágico y de una mayor síntesis de cuerpos lipídicos cuando se sobre-expresa CPT1C, aportando así información del mecanismo necesario para que CPT1C ejerza este efecto protector.
Estos estudios de CPT1C en células madre mesenquimales pueden abrir nuevas estrategias terapéuticas en el campo de la medicina regenerativa.
